Incorporation allude the town as a village when the original authority around 1831 was allowed to lapse; being reinstated around 1856 and then becoming a city around 1882. Around 1892, the Perryville, Chester, and Ste. Genevieve Railroad established rail service to Perryville followed 12 years later by the Frisco Railroad Line which laid tracks through the county and built the first railroad depot. By 1913 Perryville had its the first power plant and after money was raised by the Chamber of Commerce to build one of the first industrial spec buildings, International Shoe Company was persuaded into a Perryville plant.
